In this thesis, label-free electrical detection of protein-ligand binding using a vertical gold nanogap is presented. A sub-lithographic vertical gold nanogap was created using a sacrificial ultra-thin film deposited by atomic layer deposition (ALD) in a process similar to the formation of a cantilever in MEMS processing. Due to the atomic precision of the sacrificial $Al_2O_3$ thickness by ALD, a 7-nm nanogap was successfully fabricated. After binding streptavidin to biotin on the gold surface, a DC electrical current was measured for various DC voltages. A dramatic DC current increase was observed in the case of biotin-streptavidin binding in comparison with the other two cases: a control group filled with air in nanogap and a biotin-only binding group. There was minimal current change in the cases of the biotin-PBST group, the biotin-BSA group, and the biotin-saturated streptavidin group, as compared with the biotin-streptavidin group. At a 0.1μg/ml concentration of streptavidin (1.5 nM), the current difference before and after the protein binding was amplified by approximately 3000-fold. Also, the detection sensitivity of the vertical nanogap as the gap size varied was investigated. This arrayable, two-terminal microdevice can be used for a wide range of detection applications in the analysis of other biomolecules and for nucleic acid hybridization.
본 석사학위 논문에서는 vertical gold nanogap 소자를 이용하여 protein-ligand binding을 electrical detection 하는 방법에 대해 나타내었다. Atomic Layer Deposition (ALD) 방법에 의해 형성된 알루미나 $(Al_2O_3)$ 박막을 이용하여 전통적인 lithography의 한계를 넘는 소자를 제작하였다. ALD 방법에 의한 정확한 Al2O3 film 두께 control 에 의해 최소 7nm의 nanogap 소자를 성공적으로 제작하였다. 이 vertical gold nanogap 소자의 응용으로서 gold 전극 표면에 biotin을 고정화 시킨 후 streptavidin을 binding 하였다. 그 뒤 DC 전압을 변화 시키면서 그 전압에 따른 DC 전류를 측정하였다. 그 결과, 어떤 biomolecule도 존재하지 않는 control group (airgap)과 biotin만 고정화 된 biotin-only binding group 에 비해 biotin 과 streptavidin 이 binding 되었을 경우 급격한 전류의 증가를 관찰했다. 또한 비교실험인biotin-PBST group, biotin-BSA group, biotin-saturated streptavidin group과 biotin-streptavidin binding group으로 수행한 실험에서는 biotin-streptavidin binding group을 제외한 다른 모든 group 에서 전류 변화가 거의 없었다. 또한 streptavidin 농도가 최소 1.5nM 일 때 biotin-streptavidin이 binding 되기 전후의 전류 변화는 약 3000배 차이가 보였다. 마지막으로 nanogap size에 따른 biotin-streptavidin의 detection sensitivity를 살펴보았다. 이러한 array제작 가능하고, 두 전극을 가지는 vertical gold nanogap 소자는 다른 biomolecule의 분석이나 핵산의 hybridization을 분석하는데 널리 쓰일 것이다.
