It has been reported that the biosynthesis of sulfur containing amino acids, methionine and cysteine, might be regulated by the gene products of metJ and cysB in E. coli. In this study, metJ and cysB genes are disrupted in E. coli, respectively and investigated the change of transcriptome profile compared with that of parent W3110 strain. In metJ mutant strain, genes involved in methionine biosynthesis were found up-regulated as expected. cysB mutant showed very slow growth rate and was not able to grow in minimal medium without L-cysteine. Interestingly, genes directly involved in the cysteine biosynthesis were not significantly affected by cysB disruption. Instead, genes related to redox regulation were mainly affected. Although two mutant strains showed L-methionine overproduction and cysteine auxotrophism, respectively which are opposite effects in cellular metabolism, transcriptome profiles of two mutants largely corresponds with each other. Therefore, this study could be helpful to understand the metabolism of sulfur containing amino acids, and develop the strategy of overproduction of sulfur containing cofactors and metabolites.
For over-production of L-methionine in Escherichia coli W3110, one strategy is to redirect carbon flow from phosphoenolpyruvate (PEP) to oxaloacetate. pykA, pykF genes involved in conversion of PEP to pyruvate and ptsG gene which is involved in consumption of PEP for glucose uptake were disrupted. Resulting Escherichia coli W3110GFA strain produced L-methionine 4 times more than wild-type. However, this mutant showed increased production rate of L-phenylalanine and L-tryptophan which is a erythrose-4-phosphate derived amino acids. Simulation using MetaFlux.NET shows that, pentose phosphate pathway (PPP) of Escherichia coli W311OGFA strain is increased significantly which contribute to increase of erythrose-4-phosphate pool and hence explains the increase of L-phenylalanine and L-tryptophan production.
metJ 와 cysB 유전자 결실 돌연별이의 전사체 분석
대장균의 황아미노산 대사에 관여하는 조절 유전자인 시스테인 촉발 유전자 cysB와 메치오닌 억제 유전자 metJ 가 결실된 돌연변이주 W3110B와 W3110J를 각각 제작하였다. cysB 유전자가 결실된 대장균은 시스테인 영양 요구주 성질을 띠었지만, DNA 칩을 이용하여 전사체 분석을 하였을 때, 시스테인 생합성 관련 유전자의 전사에 주는 영향은 크지 않았다. 오히려, 산화ㆍ환원과 관계된 유전자들의 발현에 영향을 주는 것을 확인하였다. metJ 유전자가 결실된 돌연변이주에서는 메치오닌 생합성 관련 유전자의 전사가 비교적 up-regulation 영향을 받는 것을 관찰하였다. 한편 cysB와 마찬가지로 산화ㆍ환원과 관계된 유전자들의 발현이 영향 받는 것이 확인되었다. 이 결과로, cysB 와 metJ는 DNA 전사를 조절할 뿐아니라, 이 결과로 글로벌하게 미생물의 산화ㆍ환원 조절에 관여하는 것을 밝혔다.
대장균에서 메치오닌 과생산
대사공학적인 방법으로 메치오닌 과생산 균주를 제작하기 위해, phosphoenolpyruvate (PEP)에서 pyruvate로 전환되는 대사 경로와 관계된 pykA, pykF, ptsG 유전자 모두가 결실된 균주(W3110GFA)를 제조하였다. 대사 흐름 분석 기법을 이용하여 W3110GFA 균주를 시뮬레이션을 수행한 결과, TCA cycle과 pentose phosphate pathway (PPP) 대사의 흐름이 증가하면서, aspartic acid 대사 흐름 역시 증가함을 예측할 수 있었다. 아미노산 분석 결과 W3110GFA 균주에서 야생균주에 비해 L-메치오닌 생산이 약 4배 증가함을 확인하였다. 또한 L-phenylalanine과 L-tryptophan 생산도 야생 균주에 비해 2배 이상 증가하였는데, 이는 대사흐름분석 시뮬레이션 결과에 의하면, PPP 대사 흐름 증가로 인한 erythrose-4-phosphate의 pool 증가와 ptsG pykA pykF 유전자 결실에 의한 PEP pool의 증가에 의한 결과로 사료된다.
