The carboxy-terminus of the gag gene of Mason-Pfizer monkey virus (M-PMV), the prototype immunosuppressive primate type D retrovirus, encodes a 36-amino acid, proline-rich protein domain that, in the mature virion, becomes the p4 capsid protein. The p4 domain has no known role in M-PMV replication. We found that two mutants with premature termination codons that remove half or all of the p4 domain produced lower levels of stable Gag protein and of self-assembled capsids. Interestingly, yeast two-hybrid screening revealed that p4 specifically interacted with TCP-1γ, a subunit of the chaperonin TRiC (TCP-1 ring complex). TRiC is a cytosolic chaperonin that is known to be involved in both folding and subunit assembly of a variety of cellular proteins. TCP-1γ also associated with high specificity with p6, a similar proline-rich domain of human immunodeficiency virus type 1 (HIV-1). Moreover, in cells, Gag polyprotein associated with the TRiC chaperonin complex and this association depended on ATP hydrolysis. In the p4 truncation mutants, the Gag-TRiC association was greatly reduced. These results strongly suggest that the cytosolic chaperonin TRiC is involved in Gag folding and/or capsid assembly. We propose that TRiC associates transiently with nascent M-PMV Gag molecules to assist in their folding. Consequently, properly folded Gag molecules carry out the intermolecular interactions involved in self-assembly of the immature capsid.

M-PMV 복제 과정중에서 다른 부분들과는 다르게 p4 부분이 담당하는 기능은 현재까지 알려진 바가 없다. 우리가 p4 도메인의 절반 혹은 전부를 제거하는 돌연변이를 가지고 실험한 바에 의하면 이 부분의 돌연변이는 Gag 단백질의 안정성에 문제를 일으켰고, 세포내에서 캡시드를 만드는 비율도 감소시켰다. 흥미롭게도 효모a를 이용한 two-hybrid 탐색에서 p4 부분은 특이적으로 chaperonin인 TRiC의 서브유닛중 하나인 TCP-1γ와 결합했다. 이전의 실험들을 통하여 TRiC는 다양한 세포내 단백질의 구조와 서브유닛 형성에 작용하는 chaperonin이라는 것이 밝혀져 있었다. 또한 TCP-1γ는 높은 특이성을 가지고 HIV에서 유사하게 proline이 많고 Gag 중합단백질에서 C 말단에 위치한 p6 도메인과도 결합하였다. 게다가 세포내에서 Gag 중합단백질은 TRiC chaperonin과 결합하였고, 이 결합은 다른 기질들과 유사하게 ATP 분해에 의존했다. 반면에 p4 부분이 제거된 돌연변이들에서는 이 결합력이 크게 감소되었다. 이상의 결과들은 TRiC chaperonin이 Gag 단백질의 구조 및 캡시드 형성에 작용할 것이라는 강한 확신을 가지게 했다. 우리는 TRiC가 일시적으로 새로 합성된 M-PMV Gag 분자들과 결합하여 그들의 구조를 갖추는데 도움을 줄 것이라는 것을 제시한다. 결과적으로 정확하게 만들어진 Gag 단백질은 분자들간의 결합력으로 캡시드를 형성할 수 있을 것이다.
