Various types of proteinoids and lipid-conjugated proteinoids were prepared, and the potential of their applicationn in biomedical fields was investigated. Proteinoids are a novel biopolymer synthesized by anhydrous thermal condensation of naturally occurring amino acids and are known to show many properties similar to those of natural proteins. In the first part of this work, proteinoids of different compositions were synthesized, and covalent conjugation of lipids, such as phosphatidylcholine (PC) and cholesterol (Chol) to proteinoids was carried out. The structural characteristics of the synthesized proteinoids were analyzed with FRIR and $^{13}C-NMR$, and it was found that proteinoids attained non-random amino acid sequences. The conjugation of lipids was verified by the intensity increases about those peaks corresponding to the alkyl or cyclic alkyl groups, which were richly present in the lipids, from the spectra. However, when highly conjugated progeinoids were analyzed by $^{13}C-NMR$ in different solvents, namely, polar $D_2O$ and non-polar $CDCl_3$, there were striking differences in the two spectra. The ones in $D_2O$ showed peaks corresponding to the peptide chains and functional groups that are exposed to water. But, the ones in $CDCl_3$ showed almost only those peaks corresponding to cholesterol. It was suggested that Chol-conjugated proteinoids were forming aggregates in water such that the hydrophobic pendant moieties come to compact association under hydrophobic interation, and that the hydrophilic chains provide shield to the hydrophobic cores. In $CDCl_3$, the hydrophobic pendant moieties would be favorably exposed to the solvent, and the hydrophilic chains would now aggregate to compactness, instead. In addition to the structural evidences, Chol-conjugated proteinoids showed surface activity in aqueous solutions. This phenomenon is supposed to result from the two-dimensional amphiphilicity attained by the linear orientation of hydrophilic peptide chains and of cholesterol moieties at the water surface.
In the second part, observations were made on the unique amphiphilic properties of Chol-conjugated proteinoids. Chol-conjugated proteinoids dispersed in buffer solutions of various pH showed light scattering behavior that was distinctive from that of unconjugated proteinoids in a way independent of pH, and nanosized particles were detected. It was because the proteinoids hydrophobized by cholesterol-conjugation spontaneously formed aggregates in aqueous dispersions under the hydrophobic interaction of the pendant moieties. Fluorescence spectrometry was employed to investigate the microenvironment of the aggregates and the critical aggregation concentration of Chol-conjugated proteinoids, and the fluorescence spectra of pyrene indicated that the aggregate cores were highly hydrophobic. Also, the release behavior of Chol-conjugated proteinoid aggregates was studied using pyrene as a model drug, and it was found that those aggregates continuously released the loaded drug into the environment. The interactions of the proteinoid aggregates with proteins were investigated by observing the changes in the fluorescence of the tryptophan residues present in the proteins. It was suggested that the interactions of the aggregates were highly influenced by the surface charges and inherent properties of the proteins. Hydrophobized proteinoids also interacted with liposomal bilayer membranes, probably coating the liposome surfaces. They resulted in liposome-stabilizing effects by protecting the liposomal membranes from the attack of surfactant molecules. When liposomes were incubated together with lipid-conjugated proteinoids, significant size changes were observed. the hydrophobized proteinoids seemed to interact with the liposomal membranes by penetrating through the membanes.
In the last part, proteinoids containing high portions of carboxyl groups and amine groups were used in preparing a biological soft-tissue adhesive. Proteinoids were mixed with gelatin, the base material, and the whole mixture was cross-linked. Proteinoids had an enhancing effect on the bonding strength of the adhesive when the adhesive was applied onto excised rat skin. The developed adhesive nearly equals the commercial fibrin glue in bonding strength and has many advantages such as low production cost and low risk of immune problems.
다양한 종류의 프로티노이드와 지질 결합된 프로티노이드를 합성하였고, 의약학 분야로의 적용에 대한 가능성을 살펴보았다. 프로티노이드는 천연 아미노산을 무수 고온 조건에서 축합반응으로 합성하여 얻어지는 새로운 생체고분자이며, 천연 단백질과 유사한 물성을 띠는 것으로 알려져 있다. 먼저 본 연구의 첫 부분에서는 여러 가지 조성의 프로티노이드를 합성하였고 포스파티딜콜린과 콜레스테롤 등의 지질을 프로티노이드에 공유 결합시켰다. 합성된 프로티노이드의 구조적 특징은 FTIR과 탄소 NMR 등으로 분석하였는데, 이들 프로티노이드는 무작위한 아미노산 서열을 가지지 않는다는 것이 밝혀졌다. 지질 결합은 각 스펙트럼에서 지질 분자에 풍부하게 존재하는 알킬 혹은 환형 알킬 그룹에 상응하는 피크의 높이 증가로 확인되었다. 그러나, 지질 결합도가 높은 프로티노이드를 극성 용매인 $D_2O$와 비극성 용매인 $CDCl_3$에 각각 녹여서 탄소 NMR로 분석하였을 때, 두 스펙트럼이 현저한 차이를 보였다. $D_2O$에 녹인 프로티노이트 샘플은 펩티드 사슬과 관능기 등 물에 노출되는 부분에 상응하는 스펙트럼을 보였으나, $CDCl_3$에 녹인 샘플은 콜레스테롤에 해당하는 피크만 나타내 보였다. 따라서 콜레스테롤이 결합된 프로티노이드는 수상 환경에서 소수성 펜던트 기가 소수성 상호작용에 의해 응집되고 친수성 사슬은 소수성 코어의 둘레에 보호막을 형성하는 응집체를 이루는 것으로 추정하였다. $CDCl_3$에서는 소수성 펜던트 기가 용매에 자발적으로 노출되고 친수성 사슬은 오히려 안으로 응집되는 현상을 보일 것으로 추정된다. 이러한 구조적 예시 외에도, 지질 결합된 프로티노이드는 수용액에서 표면활성을 보였다. 이 현상은 물 표면에서 친수성 펩티드 사슬과 콜레스테롤 기의 방향성에서 비롯된 이차원적 양친성에서 비롯된다고 추측된다.
본 연구의 두 번째 부분에서는 콜레스테롤이 결합된 프로티노이드의 독특한 양친성에 관해 관찰하였다. 콜레스테롤이 결합된 프로티노이드를 여러 pH의 완충액에 분산시키고 광산란 거동을 관찰하였는데, 지질결합이 이루어지지 않은 프로티노이드와 달리 pH에 영향을 받지 않고 나노 크기의 입자를 형성하는 것으로 밝혀졌다. 콜레스테롤 결합으로 소수화된 프로티노이드가 수용 분산액에서 소수성 상호작용에 의해 자발적으로 응집체를 형성하기 때문이다. 형광 분석을 이용하여 콜레스테롤이 결합된 프로티노이드 코어의 미세환경과 임계 응집체 농도 등을 살펴보았다. 피렌의 형광 스펙트럼에서 프로티노이드 응집체의 코어가 매우 소수적이라는 것을 알아내었다. 또한, 피렌을 모델 약물로 이용하여 콜레스테롤이 결합된 프로티노이드 응집체의 방출 거동을 살펴보았는데, 이 응집체들은 함유한 약물을 연속적으로 방출한다는 것을 알아내었다. 프로티노이드 응집체와 단백질간의 상호작용에 대해 알아보기 위해 단백질에 존재하는 트립토판의 형광성의 변화를 관찰한 바, 응집체들의 상호작용은 단백질의 표면 저하와 단백질 고유의 기능 등에 크게 영향을 받는 것으로 나타났다. 소수화된 프로티노이드는 리포솜의 이중막과도 상호작용하는 것으로 나타났다. 프로티노이드는 리포솜 막을 코팅하여 계면활성 분자의 공격을 막아 리포솜을 안정화시키는 효과를 나타냈다. 리포솜을 지질 결합된 프로티노이드와 함께 보관하였을 때, 리포솜의 크기가 현저히 변화하는 것을 관찰하였다. 소수화된 프로티노이드는 결합된 지질 기가 리포솜 막으로 침투하여 리포솜과 상호작용하는 것으로 보인다.
본 연구의 마지막 부분에서는 카르복실 기와 아민 기를 다량 함유하는 프로티노이드를 합성하여 생체접합체를 개발하였다. 프로티노이드를 젤라틴과 혼합하여 가교시켰다. 쥐의 피부에 개발된 접합제를 적용했을 때 프로티노이드는 접합제의 접합력을 향상시키는 효과를 나타냈다. 개발된 접합제는 상용화되어 있는 피브린 접합제에 필적하는 접합력을 보였고, 낮은 생산단가와 면역 문제 저하 등 여러가지 장점을 가지고 있다.
