Kaposi's sarcoma-associated herpesvirus (KSHV) contains many cellular homologue genes, in addition to genes required for viral replication and assembly. The K9 open reading frame (ORF) of KSHV encodes a virus-encoded interferon regulatory factor (vIRF) which functions as a repressor for cellular interferon-mediated signal transduction, and as an oncogene to induce cell growth transformation. In addition, KSHV vIRF plays an important role in regulation gene expression. From genetic and biochemical analysis, we demonstrate that KSHV vIRF1 binds to a transcriptional coactivator CREB-binding protein (CBP) in vivo and in vitro. CBP plays a central role in the regulation of gene expression and is the target for several viral proteins such as adenovirus E1A, SV40 large T, and human papillomavirus E6. KSHV vIRF1 binds to the KIX domain and CH/3 region of CBP. The CH/3 region of CBP coinsides with the binding region of adenovirus E1A. We also show that vIRF1 inhibits the transactivational activity of CBP in HeLa cells. These results demonstrate that vIRF1 can modulate gene expression by inhibiting the transactivation function of coactivator CBP.
카포시 육종 헤르페스 바이러스는 복제에 필요한 유전자 이외에 다양한 세포의 유전자와 유사한 유전자들을 가지고 있다. 카포시 육종 헤르페스 바이러스의 ORF K9은 바이러스 인터페론 조절인자로 세포내에서 인터페론에 의해 유발되는 다양한 신호전달을 저해하며, 세포성장을 촉진해 종양을 일으킨다. 또한 카포시 육종 헤르페스 바이러스의 유전자 발현을 조절하는 인자로 작용한다. 생화학적인 방법을 통해 카포시 육종 헤르페스 바이러스의 인터페론 조절인자 1이 CREB-Binding protein (CBP) 과 in vivo, in vitro에서 결합한다는 것을 알 수 있었다. CBP는 세포의 유전자 발현에 중요한 기능을 하며, 따라서 많은 바이러스 단백질들의 대상이 된다. 아데노 바이러스 E1A, SV40 Tag, 파필로마 바이러스 E6들이 CBP와 결합하여 기능을 방해한다. 바이러스 인테페론 조절인자 1은 CBP의 KIX domain과 CH/3 부위에 특이적으로 결합하였다. 이러한 CBP의 CH/3 부위는 E1A에서의 경우와 일치한다. 바이러스 인테페론 조절인자 1은 HeLa 세포에서 CBP의 전사촉진 활성을 방해하였다. 이러한 결과로 볼 때 바이러스 인터페론 조절인자 1은 CBP의 전사촉진 활성을 저해함으로써 세포의 유전자 발현을 다양하게 조절할 수 있을것으로 생각된다.
