Positive strand RNA viruses constitute the major proportion of plant viruses. RNA species of plant tripartite RNA viruses whose genome is composed of three species of RNA show distinct translational activities in vitro when the virion RNA concentration is high. However, it is not known what is responsible for the differential translation of the viral RNAs. In this study, I found that cucumber mosaic virus(CMV) RNAs are also distinctly translated when the virion RNA concentration is high. In the mRNA competitive environment, RNA 4, a subgenomic RNA that encodes coat protein, showed the strongest translational activity. The competitive translated determinant in the RNA 4 was demonstrated to exist in the 5′ untranslated region(UTR) using chimeric mRNA containing viral UTR and a luciferase-coding sequence. In CMV genomic RNAs 1, 2 and 3, translation efficiencies and competitive translational activities were also investigated using chimeric CMV RNAs that contained viral UTRs and a luciferase-coding sequence. The chimeric RNAs exhibited distinct translational efficiencies and competitive translational activities. For example, the translational activity of chimeric CMV RNA 4 was about 40-fold higher than that of chimeric CMV RNA 3 in a highly competitive environment. The distinct translational activities resulted mainly from differences in competitive translational activities did not require viral proteins or 3′ UTRs. Viral 5′ UTRs alone specified the differential competitive activities. The competitive translational activities of the 5′ UTRs were as follows: RNA 4(coat protein)>RNAs 2 and 1(2a and 1a protein, or replicase)>RNA3(3a protein). In order to identify RNA features that contribute to competitive translational activity, I constructed a series of mRNA transcripts that contained RNA 4-derived, mutated 5′ UTRs that differed in stability of an optimally predicted RNA secondary structure. Stabilization of the 5′ UTR RNA secondary structure inhibited competitive translational activity. Alteration of a potential single-stranded loop to a stem by substitution mutations inhibited strongly the competitive translational activity. Tobacco plants infected with wild type virus showed a 2.5-fold higher accumulation of maximal coat protein than did plants infected with a loop-mutant virus. Amplification of viral RNA in these plants could not explain the difference in accumulation of coat protein. Phylogenetic comparison showed that potential single-stranded loops of 12 to 23 nucleotide lengths exist widely in subgroups of CMV. Since the dramatic difference in competitive translational activity between RNA 4 and vector-derived 5′ UTRs could not be explained only with the importance of a single-stranded loop region, it was investigated other RNA features that contribute to mRNA competitive translational activity. I constructed a series of mRNA transcripts that contained RNA 4-derived, mutated 5′-UTRs that differed in cap-proximal sequence context and extent of cap protrusion. The competitive translational activities of mRNA transcripts could be modulated by alterations in the cap-proximal sequence context. In contrast, cap accessibility related to the length of an unpaired cap region did not appear to be an important competitive translational determinant. The first seven nucleotides of an mRNA determined the hierachy of the competitive order. For example, the RNA 4-derived 5′ UTR was a strong competitor, while the vector-derived 5′ UTR was a week competitor. Transcripts with cap-proximal regions that contain low amounts of guanine and cytosine are common among plant RNA viruses.
대부분의 식물 바이러스는 RNA를 게놈으로 하고 있다. 이들 바이러스 중에 그 게놈이 세개의 단편으로 나뉘어져 있는 Bromoviridae에 속하는 바이러스들의 경우, 바이러스 RNA가 고농도로 존재할 때 그 번역의 활성이 차별적으로 나타나는 현상이 보고되었다. 본 연구에서, 오이 모자이크 바이러스에 있어서도 RNA가 고농도로 존재하는 상황에서 각각의 바이러스성 RNA가 차별적인 번역의 활성을 보임이 확인되었다. 고농도의 바이러스성 RNA가 존재하는 상황에서 바이러스의 껍질 단백질을 암호화하는 RNA 4가 가장 높은 활성을 보였다. 이 RNA 4의 경쟁적 번역활성이 5′ UTR에 의해 결정됨을 루시퍼라아제를 암호화하는 키메라성 RNA를 만들어 증명하였다. 또한, RNAs 1, 2 및 3에 있어서도 그 경쟁적 번역의 활성을 차별화하는 RNA 결정소가 공히 5′ UTR에 존재함도 확인하였다. 이 차별적인 경쟁적 번역의 활성을 위해 바이러스성 단백질이나 3′UTR은 요구되지 않았다. 바이러스성 RNA의 5′UTR의 경쟁적 번역의 활성의 순위는 RNA 4(껍질 단백질)>RNAs 2 및 1(RNA 복제 단백질)>RNA 3(이동 단백질)의 순서임을 확인하였다. 오이 모자이크 바이러스에 있어서 강한 경쟁적 번역의 활성을 보이는 RNA 4의 5′ UTR의 염기서열은 다소 안정적인 RNA 이차 구조를 형성할 수 있는 것으로 모델링 되었다. 전령 RNA의 번역의 활성을 저해한다는 것이 잘 알려져 있으므로, 나는 추정되는 이차구조의 안정성을 증가시키는 돌연변이 RNA들을 제작하였다. 이들의 경쟁적 번역의 활성을 조사한 결과 예상한 대로 RNA이차 구조의 안정성정도에 의존하여 그 경쟁적 번역의 활성이 조절되었다. 특히, 약 15개 정도의 염기로 구성된 추정되는 single-stranded loop 부위에 치환 돌연변이를 유도하여 stem으로 변화 시켰을 때, 그 경쟁적 번역의 활성이 강하게 저해됨을 발견하였다. 이렇게 single-stranded loop의 크기가 작아진 돌연변이성 RNA를 지닌 바이러스에 감염된 담배 식물체에서 바이러스 껍질 단백질의 최대 축적이 야생성 바이러스의 약 40% 정도의 수준이었다. 이 껍질 단백질의 축적의 차이를 바이러스성 RNA의 축적의 정도로서 설명할 수는 없었다. 이러한 single-stranded loops이 다른 많은 오이 모자이크 바이러스에 있어서도 존재할 것으로 추정할 수 있었다. 이러한 single-stranded loop은 경쟁적 번역의 활성에 관여하는 세포성 번역 개시인자의 결합 부위일 것으로 추정된다. 비록, single-stranded loop부위가 RNA 4의 경쟁적 번역의 활성에 중요한 것으로 증명되었지만, 이것만으로 플라스미드유래의 인공적 5′ UTR과 RNA 4의 5′ UTR의 경쟁적 번역에 있어서의 극적인 차별을 설명하기는 어려웠다. 따라서, 경쟁적 번역의 활성에 관여할 것으로 제시되었던 RNA 특질인 5′-Cap 구조에 대한 세포성 단백질의 접근 용이성의 영향을 알아보기 위하여, RNA 4의 5′ UTR에 있어서 Cap구조 주변의 뉴클레오티드 서열에 대해 치환, 삽입 및 결실 등의 돌연변이를 유도하였다. 그 결과 경쟁적 번역의 활성이 Cap 구조에 인접한 염기 서열의 변화에 의해 강하게 조절됨을 발견하게 되었다. 더욱이, Cap구조에 인접한 7개의 염기가 RNA 4와 플라스미드의 5′ UTR에 있어서 그 경쟁적 번역의 활성의 순위를 결정하는 결정적인 RNA 결정소임을 발견하였다. 이러한 결과는 경쟁적 번역의 활성이 Cap 구조에 인접한 뉴클레오티드 서열의 이차구조 보다는 일차구조 자체에 의해 영향을 받는 것을 의미한다. 실제로 5′ 말단에 Cap 구조를 지닌 많은 식물 RNA 바이러스들에 있어서 Cap 구조의 인접한 위치에 아데닌/우라실이 지배적으로 존재함을 발견할 수 있었다. 이를 통해 전령 RNA에 있어서 Cap 구조에 인접한 뉴클레오티드의 일차구조가 경쟁적 번역의 활성에 관여함을 밝힐 수 있었다.
