The thermo-sensitive microparticles were prepared by the spray drying method. Ethylcellulose-g-PNIPAM was synthesized by the graft copolymerization of PNIPAM onto ethylcellulose using ammonium persulphate as radical initiator in chloroform/DMSO (50/50) ml mixed solvent system. The ethylcellulose-g-PNIPAM was characterized with various techniques such as FTIR spectroscopy, NMR spectroscopy and DSC measurements. The degree of graft polymerization (DGP) which means the length of a PNIPAM grafts was calculated by the $^{13}$C NMR spectra with an assumption that the number of PNIPAM grafts (NG) is 1.
Microparticles were prepared by the spray drying method using B-191 Mini Spray Dryer. Three samples with different graft length of PNIPAM were used as the polymer and allopurinol was used as a model drug. The morphology of microparticles was studied using scanning electron microscopy(SEM). The particle size distribution of microparticles was narrow.
The release rate of allopurinol from ECGPN8 microparticles was slower at 40℃(above the LCST) than that of 25℃(below the LCST) probably due to the collapse of PNIPAM chains by temperature, while ECGPN4 microparticles didn't. The thermo-sensitive release behavior of microparticles was affected by the length of a PNIPAM grafts. A relatively large burst effect was found at the start of release and attributed to the disadvantage of the spray drying method; the rapid solvent evaporation in the spray chamber of a spray dryer results in the free drug.
The graft copolymer ethylcellulose-g-PNIPAM could be used as a component of temperature-sensitive devices in various forms and the spray drying method provides an efficient and rapid tool for the preparation of microparticles except the large burst effect.
열민감성 마이크로 입자들을 분무건조법으로 제조하였다. Chloroform/DMSO (50/50)ml 혼합용매 시스템 내에서 ammonium persulphate를 라디칼 개시제로 사용하여 ethylcellulose에 PNIPAM을 graft copolymerization 시킴으로써 Ethylcellulose-g-PNIPAM을 합성하였다. Ethylcellulose-g-PNIPAM은 FTIR spectroscopy, NMR spectroscopy, DSC 측정 등과 같은 다양한 기술들로 특성 분석되었다. PNIPAM grafts의 길이를 의미하는 degree of graft polymerization (DGP)를 number of PNIPAM grafts (NG)가 1이라는 가정 하에서 $^{13}$C NMR로 계산하였다.
B-191 Mini Spray Dryer를 사용하여 분무건조법으로 마이크로 입자들을 제조하였다. 고분자로는 PNIPAM의 graft length가 다른 세 가지 샘플들을 사용하였으며 모델 약물로는 allopurinol을 사용하였다. SEM으로 마이크로 입자들의 모양을 관찰하였다. 마이크로 입자들의 입자 크기 분포는 좁았다.
ECGPN8 마이크로 입자들로부터의 allopurinol의 방출 속도는 25℃(LCST 아래)에서보다 40℃(LCST 위)에서 더 느렸는데 아마도 PNIPAM 사슬들의 뭉침 때문으로 보인다. 반면, ECGPN4 마이크로 입자들은 열민감성 거동을 나타내지 않았다. 마이크로 입자들의 열민감성 방출 거동은 PNIPAM grafts의 길이에 영향받았다. 비교적 큰 burst effect가 방출 초기에 관찰되었는데 이것은 분무건조법의 단점-spray dryer의 spray chamber내에서의 용매의 급속한 증발로 인한 제재화되지 않은 약물의 발생- 때문에 나타난 결과로 보인다.
Graft 고분자 ethylcellulose-g-PNIPAM은 다양한 형태로 열민감성 장치의 구성성분으로 사용될 수 있었으며 분무건조법은 마이크로 입자들을 제조함에 있어 큰 burst effect를 제외하고는 효과적이고 빠른 수단이었다.
