Serratia marcescens metalloprotease inhibitor (SmaPI) is a proteinase inhibitor toward Serratia marcescens metalloprotease (SMP). As an initial step to elucidate the structure-function relationship of SmaPI, three-dimensional structure of SmaPI was computer modelled using the structure of SMP - Erwinia chrysanthemi inhibitor as template. The secondary structure elements and overall folding are very similar. To express and secrete the SmaPI in Bacillus subtilis, Bacillus expression vector, pSM704, was constructed. The DNA fragment containing the coding sequence for SmaPI was amplified and the amplified-DNA product was inserted into the downstream region of subtilisin signal sequence. The recombinant SmaPI expressed in B. subtilis DB431 was secreted into the culture medium in a large amount. After cultivation for 32 h, the amount of SmaPI secreted into the culture medium reached about 100 mg/liter when estimated by measuring inhibitory activities toward SMP. The $NH_2$-terminal amino acid sequencing analysis confirmed that authentic SmaPI and the recombinant SmaPI have the same $NH_2$-terminal amino acid sequences. The inhibitory activity of the purified recombinant SmaPI was found to be nearly equivalent to that of authentic SmaPI.
In sequential deletion analysis of the $NH_2$-terminal region of the SmaPI, SmaPIs starting at Ser-2 and Leu-3 residues, respectively, had nearly a full inhibitory activity toward SMP. However, SmaPI starting at Ala-4 residue showed severely decreased inhibitory activity. Furthermore, kinetic analysis demonstrated that SmaPI starting at the Ala-4 residue had an inhibition constant for SMP approximately 4-fold higher than that of wild-type SmaPI. The interactions of Leu-3 with SMP contribute $0.73kcal mol^{-1}$ to the overall stability of the SMP - SmaPI complex ($8.44kcal mol^{-1}$). To elucidate the detailed role of the Leu-3 residue in inhibitory activity of SmaPI, several site-directed mutations were introduced. The inhibitory activities of Leu-3 mutants in which the Leu-3 has been converted to Ala, Asp, Gly, Ile, Lys, Phe or Pro were correlated with the hydrophobicities of substituted amino acids. About $0.3kcal mol^{-1}$ is attributable to the side chain of the Leu-3 residue in the binding with SMP. From these results, it is suggested that (i) in contrast with the E. chrysanthemi inhibitor, Gly-1 and Ser-2 of SmaPI are not critical and (ii) the hydrophobicity of Leu-3 may be important in its inhibitory activity and binding with SMP.
To elucidate the role of the $NH_2$-terminal region between Ala-4 and Thr-7 in inhibitory activity of SmaPI toward SMP, several site-directed mutations were introduced. The A4R and T7A mutant SmaPIs showed a nearly full inhibitory activity. However, the inhibitory activities of Leu-5 mutants in which the Leu-5 has been converted to Ala, Gly, Ile, or Val were significantly decreased. Surprisingly, L5I and L5V mutant SmaPIs showed less inhibitory activities than L5A mutant. From these results, we suggested that inhibitory activity of SmaPI depend not the overall side-chain hydrophobicity but on the orientations and positions of respective aliphatic groups in side-chain of position 5. The side-chain of the Leu-5 residue contributes approximately $0.79kcal mol^{-1}$ to the binding of SmaPI with SMP. In other word, about 10% of the total binding energy is attributable to the side-chain of Leu-5. The inhibitory activities of Pro-6 mutants in which Pro-6 has been converted to Ala or Gly also severely decreased. The Pro-6 may have critical role in maintaining the strict conformation of $NH_2$-terminal portion that may be important in inhibitory activity of SmaPI. In conclusion, Leu-5 and Pro-6 have crucial role in inhibitory activity of SmaPI toward SMP.
To compare SmaPI with other serralysin inhibitors, E. chrysanthemi B374 inhibitor, E. chrysanthemi EC16 inhibitor, and P. aeruginosa alkaline protease inhibitor were expressed and compared with SmaPI about inhibitory activity toward SMP. Among serralysin inhibitors, SmaPI has the strongest inhibitory activity toward SMP. The specific inhibitory activities of E. chrysanthemi and P. aeruginosa alkaline protease inhibitor toward SMP were approximately 30% and 10% of that of SmaPI, respectively. From the results of chimeric protein study between SmaPI and P. aeruginosa alkaline protease inhibitor, it is suggested that the segments remote from $NH_2$-terminus may be involved in target enzyme specificity of serralysin inhibitors.
Serratia marcescens 유래의 Metalloprotease 억제제인 SmaPI는 Serratia marcescens 유래의 Metalloprotease (SMP)를 특이적으로 억제하는 단백질성 억제제이다. 이 SmaPI의 구조와 기능간의 상관성에 대한 연구를 위한 첫단계로서 이미 구조가 알려진 유사한 단백질성 억제제인 E. chrysanthemi 유래의 억제제와 SMP의 복합체의 3차구조를 기초로하여 SmaPI의 3차 구조를 computer simulation하였다. 예측된 SmaPI의 2차구조와 전체적인 단백질 접힘구조는 E. chrysanthemi 유래의 억제제와 매우 유사하였다. 이 SmaPI를 대량으로 순수하게 얻기 위하여 서브틸리신의 promoter와 분비신호를 지닌 Bacillus 발현 벡터 pSM704를 제조하였다. SmaPI를 암호화하는 유전자 서열만을 PCR을 통해 증폭한후, 이를 발현벡터 pSM704에 삽입시켜 SmaPI 발현을 위한 플라스미드 pSMPI를 제조하였다. 이 pSMPI로 형질전환된 Bacillus subtilis DB431은 상당량의 SmaPI를 세포외로 분비하였다. 배양후 약 32시간후에 세포외로 발현되는 재조합 SmaPI의 양은 SMP에 대한 억제능이의해 예측결과 약 리터당 100mg 정도에 해당하는 양이었다. 재조합 SmaPI의 N-말단 서열은 본래의 SmaPI와 일치하였으며 SMP에 대한 억제능력도 본래의 SmaPI와 거의 같음을 알수있었다.
SmaPI의 N-말단 부위의 중요성을 알아보기위해 N-말단 순차적 제거 돌연변이 기법을 수행하였다. N-말단으로부터 1개와 2개의 잔기가 제거된 변이 SmaPI의 경우, 이들은 모두 야생형 SmaPI와 거의 같은 활성을 지니고 있었으며, $K_i$ 수치도 야생형과 유사하였다. 반면, N-말단으로부터 3개의 잔기가 제거된 변이 SmaPI의 경우, 급격히 감소된 SMP 억제활성을 나타내었으며, $K_i$ 수치도 야생형의 4배 큰 수치를 가지고 있었다. 이 $K_i$ 수치로부터 SmaPI의 Leu-3 잔기가 SMP-SmaPI 복합체의 전체적인 안정성 ($8.44kcal mol^{-1}$)에 약 $0.73kcal mol^{-1}$를 기여하고 있음을 알아내었다. 이는 전체적인 안정성기여의 약 10% 정도의 수치이다. 이 SmaPI의 Leu-3 잔기의 정확한 역할을 알아보기 위해 부위특이 돌연변이 기법을 이용해 Leu-3 잔기를 Ala, Asp, Gly, Ile, Lys, Phe, 그리고 Pro으로 각각 치환하였다. 각 변이 SmaPI의 활성을 조사해본 결과, 각 치환된 잔기의 hydrophobicity와 억제활성간에 일정한 연관성이 있었다. 즉 hydrophobicity 정도와 억제활성능에 비례관계가 성립하였다. 또한 Leu-3 잔기의 side-chain만으로는 약 $0.3kcal mol^{-1}$ 정도를 전체 SMP-SmaPI 복합체의 안정성에 기여하였다. 이상의 결과로부터 (i) E. chrysanthemi 유래의 억제제에서 기대되는 것과는 달리 SmaPI의 Gly-1과 Ser-2 잔기가 억제활성에 별다른 영향을 미치지 못하며, (ii) Leu-3 잔기의 hydrophobicity가 SMP와의 결합과 SmaPI의 억제활성에 매우 중요함을 알수있었다.
SmaPI의 Ala-4와 Thr-7 잔기 사이부위의 역할을 알기위해 이부분 잔기의 부위특이 돌연변이를 도입하였다. 각 변이 SmaPI의 억제활성과 $K_i$ 수치로부터 Ala-4와 Thr-7 잔기가 억제활성에 큰 영향을 미치지 못하며, Leu-5 잔기의 경우, 전체적인 hydrophobicity보다는 side-chain을 구성하는 각 aliphatic group의 방향성과 위치가 SmaPI의 SMP 억제에 중요함을 알 수 있었다. $K_i$ 수치로부터 Leu-5 잔기의 side-chain만으로는 전체의 SMP-SmaPI 복합체의 안정성에 약 10% 정도인 $0.79kcal mol^{-1}$ 정도를 기여하였다. Pro-6 경우는 이 잔기가 SmaPI의 억제능에 중요한 SmaPI의 N-말단 부위의 특이적인 conformation 유지에 중요하다고 생각되며, 이 잔기를 Ala과 Gly으로 치환한 변이 SmaPI 모두 크게 활성이 감소하였다.
SmaPI와 다른 serralysin 억제제를 비교하기위해, E. chrysanthemi B374 inhibitor, E. chrysanthemi EC16 inhibitor, 그리고 P. aeruginosa alkaline protease inhibitor를 Bacillus subtilis에서 발현, 정제하였다. SMP에 대한 이들의 억제능 측정결과 SamPI가 가장 SMP를 강하게 억제하였다. 융합 재조합 단백질 실험 결과, SmaPI의 N-말단 이외의 부위는 억제제의 specificity를 부여하는데 관여하는 것으로 사료된다. 이상의 연구를 통해서 Metalloprotease와 그 억제제의 일반적인 억제, 결합 방식을 알아내는데 기초적인 정보를 제공하리라 사료된다.
