Cytotoxicity and apoptosis mediated by selenium were investigated in human promyelocytic leukemia (HL-60) cells. Treatment of selenite or selenodiglutathione (SDG), an initial metabolite of selenite, resulted in concentration-dependent cytotoxicity, measured by lactate dehydrogenase (LDH) leakage assay for detection of membrane damage, and by tetrazolium salt reduction assay reflecting mitochondrial function within the cell. Treatment with selenite or SDG also induced apoptosis in a dose-dependent manner, as detected by enzyme-linked immunosorbent assay (ELISA) specific for histone-associated DNA fragments, DNA fragmentation assay, and gel electrophoresis. SDG exerted more potent effect on the induction of apoptosis and cytotoxicity. Time-course study of cellular selenium uptake suggests that the stronger effect of SDG is largely due to faster and greater selenium uptake rate. The activity of poly(ADP-ribose)polymerase (PARP) was increased preceding DNA fragmentation, suggesting an involvement of PARP in selenium-mediated apoptosis. The dose-response data of apoptosis induced by selenite or SDG were similar to those of cytotoxicity, implicating a relationship between the induction of apoptosis and cytotoxicity. Zn, an inhibitor of apoptosis, dose-dependently blocked not only the induction of apoptosis, but also the membrane damage induced by selenium, corroborating this hypothesis. It was noted that the inhibition of apoptosis by Zn exerted little protective effect on cytotoxicity at higher concentrations of selenium, compared with a perfect protective effect at low concentration of selenium. These results suggest that cytotoxicity induced by selenium may be partially correlated with apoptosis. Treatment with cyanide, a specific trapper of SDG, was effective to reduce cytotoxicity and apoptosis induced by selenite or SDG in a dose dependent manner. The similar trends of reduced cytotoxicity and apoptosis were shown by co-treatment with $Hg^{2+}$, which has been known to be a specific trapper of selenide, the final reduced form of selenite. These results suggest that the reduction of selenite to selenide may be crucial for the induction of apoptosis and cytotoxicity in the cells. Treatment with $Hg^{2+}$ also blocked apoptosis mediated by methylselenocysteine (MSC), implicating that the conversion of MSC to selenide is crucial for the mechanism by MSC induces apoptosis. These results suggest a possibility that selenide is a universal key intermediate of the mechanism by which any form of selenium induces apoptosis.
인간 미분화 골수암 (HL-60) 세포주에서 셀레늄에 의해 유도되는 세포 예정사와 세포 독성을 연구하였다. 세포 독성은 LDH leakage assay와 WST-1 assay로 검출하였고, 세포 예정사는 주로 DNA-histone 절편에 대한 ELISA와 DNA fragmentation assay로 정량하였다. Selenite와 selenite의 환원 대사물인 selenodiglutathione (SDG)는 HL-60 세포의 세포 예정사와 세포 독성을 농도 의존적으로 유도함이 관찰되었다. SDG가 selenite에 비해 더 높은 효과를 나타내었는데, 이는 SDG의 세포 내 유입율이 상대적으로 더 높은 것에 기인하는 것으로 보인다. 셀레늄에 의해 유도되는 DNA의 절편화가 일어나기 전에 poly(ADP-ribose) polymerase의 활성이 증가함이 관찰됨으로써, 이 효소가 셀레늄에 의해 유도되는 세포 예정사 기작에 간여할 것으로 예견된다. 세포 예정사의 저해제인 Zn는 셀레늄에 의해 유도되는 세포 예정사를 저해함과 더불어 세포 독성을 부분적으로 억제함으로써, 셀레늄에 의한 세포 독성이 부분적으로 세포 예정사 기작에 기인함을 암시하였다. SDG와 특이적으로 반응하는 cyanide는 selenite와 SDG에 의해 유도되는 세포 독성과 세포 예정사를 억제함이 관찰되었다. Selenite의 환원 대사 과정의 마지막 산물인 selenide와 특이적으로 결합하는 것으로 알려진 $Hg^{2+}$ 역시 이 기작을 강력히 저해하였다. 이는 selenite의 환원 대사에 의한 selenide의 생성이 selenite에 의해 유도되는 세포 예정사에 필수적인 과정임을 나타낸다. 또한, 셀레늄의 대사 경로 중 환원 대사를 거치지 않고 methylselenol의 형태로 직접 대사되는 것으로 알려진 methyselenocysteine에 의해 유도되는 세포 예정사도 $Hg^{2+}$에 의해 억제되었다. 이러한 결과를 바탕으로, 셀레늄에 의해 유도되는 세포 예정사의 기작에서 공통적으로 selenide가 필수적인 중간 대사물로 작용하리라는 가설을 제시한다.
