It has been shown that most proteins interact with lipid bilayer strongly, only when converted into a partially unfolded structure such as molten globule. SecA protein, one of the translocation machinery of Escherichia coli, is an exceptional water soluble protein which native form can readily penetrate the membrane and lipid bilayer under physiological condition. In order to see whether the native SecA exhibits the partially unfolded characteristics, temperature- and denaturant-induced denaturation studies of SecA performed by following the changes of the mean residue ellipticity of the far-UV circular dichroism and the intensities of tryptophan and 8-anilinonaphthalene-1-sulfonic acid (ANS). Solvent accessibility of SecA was also investigated using the techniques such as the hydrogen-tritium exchange kinetics of amide-backbone hydrogens, ANS binding and quenching of tryptophan fluorescence by iodide. It appears that denaturation of SecA by heat and denaturant goes through intermediates with the characteristics of molten globule state. The solvent accessibility studies show that 60% of backbone hydrogens of SecA is exchangeable, while 75% of tryptophans are exposed to the solvent. These results are compared with the properties of native and molten globule forms of -lactalbumin (-LA) as well as those of apocytochrome c. The exposure of hydrophobic residues as judged from the ANS binding and the extent of amide hydrogen exchange of SecA were comparable to those of native -LA. On the other hand, equilibrium unfolding experiments showed that SecA is less stable than native -LA. The stable N-terminal 45-kDa fragment caused by the tryptic digestion in aqueous solution, the change of endogenous ATPase activity by guanidine-hydrochloride and the extent of quenching of Trp fluorescence which is concentrated in the C-terminal end are suggestive of the C-terminal half of SecA being more flexible than the rest of the protein. The overall conclusion is that the SecA, as a whole, has lower conformational stability and somewhat open structure due to relatively unstable C-domain which may facilitate its penetration into lipid bilayer.
많은 단백질은 molten globule과 같은 부분적으로 풀린 구조로 바뀔 때 지질 이중층과 강하게 결합하는 것으로 알려져 있다. 이 molten globule 구조는 인위적으로는 산성 환경, 높은 온도등에서 형성되고, 단백질 합성 도중의 구조 형성 과정이나 산성 인지질막 근처와 같은 자연 조건에서도 형성된다. 단백질의 molten globule 구조는 단백질의 원래 구조에 비해 2차구조는 유사하지만 3차구조가 거의 파괴되어 단백질의 내부, 특히, 소수성 부분이 노출되어 있다. 이 때문에 molten globule은 인지질막에 쉽게 결합하는 것으로 알려져 있다.
대장균의 막전위 과정에 관여하는 단백질로 ATPase 활성을 가지고 있는 SecA는 생리적 조건하에서 생체막과 지질이중층을 쉽게 통과할 수 있는 특이한 수용성 단백질이다. 이러한 성질은 SecA가 부분적으로 풀린 구조 또는 molten globule 구조를 가지기 때문으로 생각되어, 구조의 안정성과 구조에 대한 용매 접근성 측정을 통하여 SecA 구조 특성을 관찰하였다. 안정성 평가를 위해, SecA의 구조를 온도와 구조변성제로 풀면서, 2차구조의 안정성은 far-UV CD의 신호 변화로, 3차구조의 안정성은 tryptophan 잔기와 ANS의 형광 변화로 관찰하였다. 2차구조는 3차구조 보다 안정하였고, SecA는 구조변화 과정에서 molten globule 특성을 갖는 중간체를 형성하였다. 구조변성제에 의한 구조변성으로 부터 SecA의 구조 안정성에 대한 자유에너지 변화를 계산하였고, 생리적인 환경에서도 SecA는 다른 단백질들보다 불안정 하였다. SecA에 대한 용매의 접근성은 단백질 골격 수소의 삼중수소 치환 동역학, ANS 결합, 요오드 이온에 의한 tryptophan 형광의 감소량 측정으로 관찰하였다. SecA의 골격 수소중에서 60%는 빠르게 치환 가능하고, 나머지 수소는 치환 속도가 아주 느린 것으로 관찰되었다. 즉, SecA 구조의 일부는 유동적이고, 나머지 부분은 상대적으로 안정한 것으로 생각된다. SecA에는 친화력이 다른 두 종류의 ANS 결합 장소들이 존재하고 많은 소수성 부분과 tryptophan의 75%가 용매에 노출되어 있다는 것을 알 수 있었다. 이 결과들을 alpha-lactalbumin (alpha-LA)과 apocytochrome c의 구조 특성과 비교하였다. alpha-LA는 산성 환경에서 molten globule 구조를 형성하고 인지질막에 쉽게 결합한다. apocytochrome c는 정상적인 환경에서도 쉽게 산성 인지질 막에 결합하고, 막을 통과하는 SecA와 유사한 특성을 갖는다. SecA의 소수성 잔기들의 노출과 수소 교환의 정도는 alpha-LA의 특성과 비슷하였다. 반면, 평형 구조변성 실험은 SecA가 alpha-LA보다 안정성이 낮다는 것을 보여주었다. 수용액에서 trypsin 분해 과정에서 남은 상대적으로 안정한 N-말단 45-kDa 단편과 Guanidine-hydrochloride에 의한 SecA 고유 ATPase 활성의 변화는 SecA의 N-말단 보다 C-말단 쪽이 더 유동적이라는 것을 암시한다. 결론적으로, SecA의 구조는 안정성이 낮고, SecA는 구조변성 과정에서 molten globule과 유사한 중간체를 형성하며, 상대적으로 불안정한 C-domain에 의해 SecA는 다소 풀린 구조를 갖는다.
