Two-stage electrodialysis was studied for its application to lactic acid recovery from fermentation broth. Suitable ion exchange membranes for this purpose were selected by using a newly designed membrane-fouling test procedure, and then with the selected membranes two-stage electrodialysis was carried out: recovery of sodium lactate from fermentation broth (desalting electrodialysis) and acidification of the purified sodium lactate (water-splitting electrodialysis). Membrane-fouling tests were carried out with a 0.5 N NaCl solution, model solutions, and fermentation broth. Among the membranes tested(Tokuyama Co., Japan), CMS, CMB and CM-1 as cation exchange membranes, and AM-1 as anion exchange membrane showed relatively low electrical resistances to be selected.
Desalting electrodialysis was operated in a constant-current and a subsequent constant-voltage modes, and the switching point was predetermined from limiting current density measurement data. The limiting current density was proportional to lactate concentration up to 10 g/L for the combination of CM-1 and AM-1, but it could not be found beyond this level. The relationship between the electrical resistance of membrane stack and the lactate concentration was identified. The amount of water transferred due to electroosmosis which caused volume changes in the feed and permeate solution was experimentally determined also. For three possible combinations of cation exchange and anion exchange membranes selected by the fouling tests, desalting electrodialysis was carried out with fermentation broth. The electrodialyzer consisting of CMS and AM-1 showed the most desirable performance in terms of glucose rejection and hardness removal. The lactate recovery and the glucose rejection were as high as 99% and 84%, respectively, and the magnesium and calcium ion rejection were as high as 71% and 44%, respectively. Such low rejections of magnesium and calsium ions suggested that an additional ion exchange step would be required to completely remove those divalent ions which otherwise could cause serious membrane fouling problems in the subsequent water-splitting electrodialysis. The current efficiency was about 97%, and the energy consumption for the recovery of 1 kg of lactic-acid-equivalent was about 0.27 kWh. Similar experimental results were obtained for model solutions also (data not shown).
Water-splitting electrodialysis to convert sodium lactate to lactic acid and to recover sodium hydroxide was operated only in a constant-current mode. In the case of model solutions, the recovery of sodium hydroxide was in the range of 88 to 93%. The current efficiency was 81 - 84 % and the energy consumption for the recovery of 1kg of lactic acid was 0.54 - 0.71 kWh. In the case of fermentation broth, the recovery of sodium hydroxide was in the range of 94 to 97%. The current efficiency was 77 - 84% and the energy consumption for the recovery of 1kg of lactic acid was 0.54 kWh.
Based on the experimental results with model solutions, mathematical models were developed for both of desalting and water-splitting electrodialyses, in which time-changes in the feed and permeate volumes and the electrical resistance were considered. Model predictions of lactate concentration, volume changes, switching time, and energy consumption were in good agreement with the experimental data. The prediction of total operating time for desalting electrodialysis showed some errors. However, it was considered to be due to difficulties involved in determinating the termination time in actual operation.
One-stage electrodialysis which has a potential of cost reduction and process simplification was studied. In this process, each electrodialyzer cell consisted of a bipolar(BP-1, Tokuyama Co.) and an anion exchange membranes, and sodium lactate recovery and acidification occurred simultaneously. Three different anion exchange membranes were tested : AM-1 selected through the fouling tests, ACM with a characteristics of low proton transport, and AFN (Tokuyama Co.) with characteristics of diffusion membrane. Among these membranes, AFN showd the best performance, that is, low energy consumption of 1.38 kWh and high current efficiency of 38.6 %. However, one-stage electrodialysis consumed more energy than the two-stage electrodialysis. Therefore, it was considered to be unfeasible in the economic point of view to be applied for lactic acid recovery.
A hybrid process of nanofiltration and water-splitting electrodialysis was investigated as an alternative to the two-stage process for the purpose of elimination or at least minimization of the necessity of the ion exchange step. Two kinds of nanomembranes were tested; NTR-729(plate type, Nitto-Denko Co.) and NF-45 (Sprial type, Dow Co.). To determine the optimal operating conditions of nanofiltration, effects of pressure, lactate concentration, pH, and impurities were studied. Under conditions of high pressure, high lactate concentration, low impurity contents, and low pH, lactate rejection was as low as 10%, and flux was as high as 3.5 L/㎡h for NTR-729 and model solutions. The rejection of glucose and protein were 24~27%, and 80%, respectively. Magnesium rejection was about 97%. In the case of NF-45 with fermentation broth, lactate rejection was lower than 21% while the rejection of acetate, a major byproduct, was higher than 27%. Magnesium rejection was 78~91% and calcium rejection was 65~83%, while they were lower than 71% and 44%, respectively in desalting electrodialysis as mentioned earlier. In the subsequent water-splitting electrodialysis, both sodium lactate conversion to lactic acid and sodium hydroxide recovery were about 96%.
식품산업에서 널리 사용되던 유산은 최근 환경문제와 관련하여 새로운 용도로 관심이 모아지고 있다. 생분해성 고분자인 poly(lactic acid)의 원료로서뿐 아니라 green solvent, 화학합성의 원료로서 앞으로 그 중요성이 커질 것으로 기대된다. 유산은 화학합성에 의한 방법과 발효에 의한 방법으로 현재 생산되고 있다. 화학합성에 의한 방법은 라세미 화합물을 얻는데 반하여 발효에 의한 방법은 원하는 이성질체만을 생산하기 때문에 발효에 의해 생산된 유산은 의료용, 식품용 등으로 폭넓게 적용이 가능하다. 기존의 발효에 의한 유산생산공정은 부산물 발생, 고순도의 유산 생산을 위한 복잡한 공정, 비싼 생산비의 문제점 등을 가지고 있다. 따라서, 경제적으로 유산을 생산하기위해 복잡한 정제공정을 개선하고자 하였다.
전기투석은 이온교환막을 통해 이온을 선별적으로 통과시킴으로서 이온교환 또는 분리ㆍ정제를 하는 공정이다. 빠른 처리, 비이온성 물질의 효과적인 제거, 생산물의 농축이 장점이고 무엇보다도 부산물 생성이 없다는 게 전기투석공정의 가장 큰 특징이다. 본 연구의 목적은 전기투석을 이용한 발효액으로부터의 유산 정제, 공정 최적화를 위한 공정 모델링, 그리고 유산정제를 위한 새로운 공정의 개발이다. 유산 정제를 위해 2단계 전기투석 공정, 1단계 전기투석공정, 나노여과 및 물분해전기투석공정을 비교 실험하였고, 전기투석공정의 최적화를 위해 공정 모델링을 수행하였다.
전기투석을 이용하여 유산을 분리ㆍ정제하는데 적당한 이온교환막을 선택하기 위한 fouling test방법을 제안하였는데 이 방법을 이용하면 여러 가지 종류의 이온교환막을 같은 조건에서 비교할 수 있으며 전기적 저항이 작은 막을 선별할 수 있다. 이 방법을 이용하여 모델 용액 및 발효액을 가지고 양이온, 음이온 교환막 각각 5종류를 비교한 결과, 양이온 교환막으로는 CM-1, CMB, CMS 세가지 막이 가장 낮은 전기저항을 보였으며 음이온 교환막으로는 AM-1의 성능이 가장 뛰어났다. AM-1의 음이온 교환막과 세가지 양이온교환막을 이용하여 탈염 전기투석을 한 결과 유산의 회수율은 약 98%를 보였고, 1kg의 유산을 정제하는데 드는 에너지 소비량도 약 0.27 kWh로서 세가지의 양이온교환막에 대해서 거의 비슷한 값을 보였다. 그러나, 포도당과 2가 금속이온의 제거율에서 CMS가 다른 두 이온 교환막 보다 좋은 성능을 보였다.
탈염과 물분해 전기투석으로 구성된 2단계 전기투석을 이용한 유산의 분리ㆍ정제에 대해 연구하였다. 탈염전기투석은 한계전류밀도를 보이는 유산 농도에서는 일정 전압 방식을, 한계전류밀도를 나타내지 않는 유산 농도에서는 일정 전류 방식의 조업 방법을 택하여 조업하였다. 한계전류밀도는 유산 농도에 비례하여 증가하였고, 유산 농도가 10g/L 이상일 때에는 한계전류밀도를 관찰할 수 없었다. 유산 농도변화에 따른 전기투석장치에 걸리는 저항변화를 관찰하였고, 전류에 따라 저항과 농도사이의 관계가 변한다는 사실도 밝혔다. 전기투석 후 변화된 feed와 permeate에서의 부피변화를 관찰하였고 부피변화는 이동한 유산의 hydration number와 관련이 있음을 밝혔다. 실험으로부터 구해진 Na이온과 Na-lactate의 hydration number는 각각 2.4와 15.8였다. 모델 용액을 이용하여 부피변화, 유산 농도에 따른 저항변화, 한계전류밀도 등의 실험결과를 바탕으로 탈염 및 물분해 전기투석 공정에 대한 수학적 모델을 제시하였다. 모델에서 제시한 feed와 permeate에서 유산의 농도변화, 부피변화, switching 시점, 에너지 소비량 등의 예측값은 실험값과 일치하였다. 총조업시간의 예측값은 모델값과 약간의 오차를 보였는데, 그 이유는 조업 후반부에 유산의 농도가 매우 느리게 변하기 때문에 실험 종료시간을 정확히 맞추기 힘들기 때문으로 실제적인 공정에서는 큰 문제가 없을 것으로 보인다. 2단계 전기투석을 이용하여 발효액으로부터 98 % 이상의 유산을 회수할 수 있었고, 1kg 유산을 정제하는데 총 0.81 kWh의 에너지 소비량을 보였다.
2단계 전기투석을 이용하여 유산과 가성소다의 동시회수보다 유산만의 회수를 목적으로 1단계 전기투석공정을 제안하였다. 앞의 fouling test에 의해 선정된 AM-1과 diffusion과 이온 교환막의 성질을 동시에 지니고 있는 AFN, $H^+$이온에 대한 낮은 투과율을 보이는 ACM 등의 세가지 음이온교환막을 이용하여 모델용액 및 발효액으로부터 유산을 정제하였다. 세가지 음이온교환막중 AFN이 가장 좋은 성능을 보였으나, 유산의 회수율뿐 아니라 에너지 소비면에서 2단계 전기투석 보다 훨씬 뒤떨어지는 결과를 보였다. 그 이유는 음이온 교환막에 대한 $H^+$이온의 투과율이 매우 높기 때문에 음이온교환막을 통과하는 도중이나 그전에 유산의 중성화가 일어나 단순한 확산으로 유산이 통과하기 때문인 것으로 보인다. 따라서, $H^+$이온의 확산을 완전히 막는 음이온교환막이 개발되지않으면 1단계 전기투석은 현실성이 없는 것으로 판단된다.
2단계 전기투석법을 대체하기위한 나노여과와 물분해 전기투석을 이용한 새로운 유산 정제법을 제안하였다. 2단계 전기투석공정은 2가 양이온에 의한 막의 손상을 피하기 위해 탈염 전기투석과 물분해 전기투석 사이에 이온교환크로마토그래피 공정이 필요한데 탈염 전기투석과 이온크로마토그래피 공정을 하나의 공정으로 처리할 수 있는 공정이 나노여과이다. 나노여과의 최적공정조건을 결정하기위해 압력, 유산 농도, pH, 불순물 등의 영향을 살펴보았다. 압력이 높을수록, 유산 농도가 높을수록, pH가 낮을수록 낮은 유산 rejection(10%)과 높은 유량을 보였다. 불순물로서 포도당의 rejection은 24~27%를 보였고 단백질의 rejection은 약 80%를 보였다. 발효액을 가지고 실험한 결과 부산물인 아세트산의 rejection은 약 27%, 2가 양이온인 $Mg^++$, $Ca^++$의 rejection은 각각 78~91%와 65~83%를 보인 반면 유산의 rejection은 21% 이하로 낮은 값을 보였다. 나노 여과한 유산용액을 물분해 전기투석하여 약 96%의 유산 전환율과 가성소다 회수율을 보였다.
