Lipases and proteases from different commercial sources were screened for their ability to catalyze the transesterification of glucose and activated N-blocked phenylalanine, tBoc-L-Phe-COOTFE. A commercial protease from Bacillus licheniformis, Optimase M-440,was found to be most effective for this purpose. The enzyme showed a broad substrate specificity toward various monosaccharides. Various monosaccharides and their derivatives(hexosamines, glycosides and sugar alcohols) were effectively acylated with tBoc-L-Phe-COOTFE by Optimase M-440 in pyridine.
On a basis of $^13C$-NMR analysis, glucose was acylated only at the C-6 position. No di- and triesters were generated because of steric hindrance. D-amino acids also can be incorporated effectively into carbohydrates with Optimase M-440. Various solvents were tested as a reaction medium for carbohydrate acylation study. Hydrophobicity of organic solvents was shown to be independent of the transesterification activity of Optimase M-440. Disaccharides were also shown to be acylated as effectively as monosaccharides with amino acid esters. Acylation of disaccharides with activated amino acid ester, tBoc-L-Phe-COOTFE resulted in the formation of diesters, which are appropriate monomers for polymerization.
Various disaccharides could be acylated with amino acid ester in pyridine. Solubility of disaccharides in pyridine could be judged from the solubility data in tert-butanol and melting points. Bulky carbohydrates such as raffinose could be also nucleophiles in the Optimase M-440 catalyzed acylation.
$^13C$ NMR data enabled us to assign structures to the mono- and diester products with the sucrose monoester identified 1'-O-, 6-O-, and 6'-O-monoesters and diesters identified as 1',6-O- and 1',6'-O-diesters. In case of trehalose, the monoesters were identified as 6-O- and 3'-O-monoesters and diester products identified as 6,6'-O- and 6,3'-O-diesters
Analysis of acylation site indicated sucrose was acylated at primary hydroxyls and no triesters of sucrose was detected. Primary hydroxyls were acylated without distinct preference. Trehalose of two glucose units was acylated at one primary hydroxyl(6-OH) and one secondary(3-OH). 6-OH was preferentially acylated than 3-OH.
Optimization of enzymatic synthesis of sucrose diesters as monomers for synthesis of biodegradable polymers was performed in terms of temperature, lyophilization of enzymes with lyoprotectants, TFE content, mole ratio, water content, water activity, presence of molecular sieves, kind of esters and acyl donors. The optimal temperature was 45 in terms of enzyme stability and solubility of substrates. Lyophilization with sucrose stabilized Optimase M-440 from reversible denaturation. Initial TFE do not have much effect on enzyme activity. Water activity affects the transesterification activity even though water is not involved directly in the reaction. The optimal water activity was 0.2-0.3. The addition of molecular sieve had an adverse effect probably because it stripped out 'essential water' from the enzyme. Various esters were synthesized and tested as acyl donors. TFE, CM, Oxime esters gave more than 40% of conversion of sucrose in 12hr. Free acid, methyl, and ethyl ester gave much lower conversion(less than 5%).
Hydrophobic amino acids such as phenylalanine, leucine and methionine gave higher reaction rate and conversion(over 50% in 24h) compared to less than 30% conversion of basic(lysine) and acidic(aspartic acid) residues. Even though tyrosine is of hydrophobic nature, much lower reactivity than phenylalanine was observed because of -OH of aromatic ring.
Sucrose 1'6-O-diesters and trehalose 6,6'-O-diesters were selected for the further polymerization study. They were successfully treated with TFA for removal of tBoc group to generate two amine functionalities available for polycondensation reactions. The optimization of cleavage reaction time revealed 30min is enough for complete removal. The glycoside bonds were stable under these conditions, confirmed by MALDI-TOF mass spectrometry. Chemical polymerization of N-deblocked sucrose 1',6-O-diester with TCE-glutarate with zinc acetate as a catalyst gave polymers of molecular weight of 15,000 determined by MALDI-TOF MS.
Enzymatic chemoselective acylation of glucosamine gave C-6 acylated products, which was confirmed by $^{13}C$ NMR analysis. Nucleotides were shown to be effectively acylated with Optimase M-440 in anhydrous pyridine, confirming broad substrate specificity of the enzyme. Thymidine and uridine gave higher conversion of amino acid esters because they have higher solubility in organic solvents. These enzymatically synthesized amino acid-sugar conjugates with glucosamine and nucleotides can be used in the chemical synthesis of polymers which are useful as polymeric drug conjugates for chemotherapy.
Chemoenzymatic synthesis of polymers represents a significant advantage over either purely chemical approaches(enhanced regioselectivity) or purely enzymatic routes(higher reaction rate and substantially higher Mw). This work represents another advance in the use of combined enzymatic and chemical synthesis in nonaqueous media. The methodology described in this work can be applied to efficient and selective chemoenzymatic production of sugar containing polymers without multistep chemical procedures.
효소의 뛰어난 위치 선택성(regioselectivity)를 이용하여 당과 아미노산이 함유된 glycoconjugate의 효소적 합성에 대한 연구를 수행하고 이를 이용하여 고분자반응의 단량체로 이용하고자 하는 연구가 수행되었다. 상업적으로 판매되는 lipase와 protease들에 대하여 단당류인 포도당과 아미노기가 보호된 활성화된 아미노산 에스터인 tBoc-L-Phe-COOTFE의 transesterification반응의 촉매능력이 조사되었다. Pyridine을 반응용매로 하여 screening한 결과 Bacillus licheniformis에서 유래하는 Optimase M-440이라는 효소가 매우 효과적인 효소임이 발견되었다. 이 효소는 여러 가지 단당류나 그 유도체들(hexoamines, glycosides, 당알콜)에 대해서도 넓은 범위의 기질 특이성을 나타내었다. $^13C$-NMR 분석을 통해 포도당의 C-6의 위치에 아미노산이 도입된다는 것이 확인되었고 diester나 triester는 입체적 장애로 인해 생성되지 않았다. D-아미노산도 L- 아미노산에 비하여 20%정도의 전활율로 Optimase M-440에 의해 당에 도입될 수 있음이 관찰되었으며, 무수 유기용매하에서 transesterification반응을 수행한 결과 pyridine에서 가장 높은 활성을 보였으며 THF나 tert-amyl alcohol도 반응용매로 사용할 수 있었다. 단당류인 포도당의 경우 아미노산이 하나가 도입됨으로 고분자반응의 단량체로 이용할 수 없으므로 이당류에 대한 아실화 반응을 수행하였다.
이당류도 단당류와 같이 아미노산에 의한 아실화 반응이 이루어짐이 관찰되었다. 이당류의 경우에는 고분자반응의 monomer로 쓰일 수 있는 diester가 합성되었다. 25℃에서 55℃까지의 범위에서 45℃가 반응속도와 효소의 활성면에서 선택되었다. Pyridine에서의 이당류의 용해도는 이당류의 녹는점과 상관관계가 있음이 관측되었다. Raffinose와 같은 삼당류도 아실화반응의 nucleophile이 될 수 있는 것으로 볼 때 Optimase M-440은 넓은 기질 특이성을 가지는 것으로 나타났다.
$^13C$ NMR분석을 통해 sucrose와 trehalose의 위치선택성을 고찰하였다. Sucrose는 1'-O-, 6-O-, and 6'-O-monoester와 1',6-O- and 1',6'-O-diester가 합성되었고 triester는 합성되지 않았다. Trehalose의 경우 6-O- and 3'-O-monoester와 6,6'-O- and 6,3'-O- diester가 얻어졌으며 역시 입체장애로 인해 triester는 합성되지 않았다. Sucrose는 primary위치의 3개의 hydroxyl?L}, trehalose의 경우는 하나의 primary hydroxyl과 하나의 secondary hydroxyl에 아미노산이 도입되었다. 고분자반응의 단량체로서의 Sucrose diester의 합성을 위해 온도, 동결건조, trifluoroethanol의 저해, 몰비율, 물의 양, 물의 활성도, molecular sieve, 아미노산의 종류, 아미노산 ester의 종류별로 최적화 실험을 수행하였다.
초기의 trifluoroethanol은 효소의 활성에 별 영향을 끼치지 못하였고, 물을 초기에 첨가한 경우에는 기질중에 하나인 아미노산 에스터를 가수분해함으로 sucrose diester의 생성이 감소하였다. 그러나 효소를 물의 활성도가 0.2-0.3으로 유지한 뒤 유기용매에 도입하면 반응속도가 2배 정도 증가되었다. 아미노산 에스터 중에서는 TFE, cyanomethyl, oxime ester가 12시간 반응에 40%의 sucrose 전환율을 보였으나 free -COOH, methyl, ethyl ester의 경우 5% 이하였다.
phenylalanine, leucine, methionine는 24시간에 50%의 sucrose전환율을 보였으나 측쇄가 큰 아미노산인 lysine, aspartic acid의 경우는 30%이하였다.
효소로 합성된 sucrose 1'6-O-diester와 trehalose 6,6'-O-diester를 고분자반응의 단량체로 사용하기 위해서 보호기로 도입된 tBoc을 trifluoroacetic acid로 처리하였다. 반응시간을 30분으로 줄임으로써 이당류내에 존재하는 glycosidic bond를 그대로 유지할 수 있음을 MALDI-TOF 질량분석을 통해 확인하였다.
-$NH_2$를 2개 가지는 당과 아미노산의 glycoconjugate와 diacid를 이용하여 축중합반응을 수행하였다. Glutaric chloride와 trehalose 6,6'-O-diester를 사용하여 계면중합한 경우에는 분자량이 3300으로 나타났는데 MALDI- TOF질량분석을 통해 당에 미반응으로 남아 있는 -OH에 반응하는 것으로 측정되었다. TCE-glutarate를 diacid로 사용하고 sucrose 1'6-O-diester를 아민화합물로, zinc acetate를 촉매로 THF에서 축중합한 경우 MALDI-TOF질량분석을 통해 분자량이15000정도의 고분자가 합성되었음을 관측하였다.
당에 아민(-$NH_2$)기를 이미 한 개 가지고 있는 glucosamine의 경우 Optimase M-440을 이용하여 아미노산이 C-6에 도입됨을 $^13C$ NMR분석을 통해 알 수 있었다. Thymidine, uridine등과 같은 핵산들로 아미노산 에스터에 의해 같은 효소로 아실화 반응을 수행하였다. 이와 같은 아미노산-당 복합체들도 제약을 붙여 chemotherapy에 사용할 수 있는 고분자 합성의 단량체로 사용할 수 있다.
