Brevibacterium ammoniagenes and related bacteria are important for the industrial production of the flavor-enhancing nucleotides, inosine-5'- monophosphate (IMP) and guanosine-5'-monophosphate (GMP). Guanine analogue, 6'-mercaptoguanine (MG), can strongly inhibit the biosynthesis of purine ribonucleotides and act as a "pseudofeedback inhibitor". MG-resistant mutants have been obtained by loss of the IMP-GMP pyrophosphorylase activity with a decreased capacity to form MG ribonucleotide. Br. ammoniagenes can convert MG to its nucleotide. Therefore, it followed that increased IMP production, by conferring MG-resistance, may be due to the release of the major regulatory enzyme of purine biosynthetic pathway, 5'-phosphoribosyl-1'- pyrophosphate (PRPP):glutamine amidotransferase (EC2.4.2.14) (hereafter PRPP amidotransferase),from feedback inhibition. Thus, we have attempted to clone the gene encoding PRPP amidotransferase of Br. ammoniagenes by cloning the DNA fragment conferring MG-resistance to the wild type strain Br. ammoniagenes ATCC6872. In the current study, 6'-mercaptoguanosine (MGS) was used as analogue other than MG because of its good solubility in water. PRPP amidotransferase has been studied in many organisms and genes or cDNA sequences encoding this enzyme have been cloned from many organisms.
IMP-overproducing strain Br. ammoniagenes IPR-1 was constructed by conferring MGS-resistance. From the IPR-1 strain, DNA fragment conferring MGS-resistance to the wild-type strain ATCC6872 was cloned and its nucleotide sequence was determined. Several ORFs were identified in the sequence and one of the ORFs showed high degree of homology with genes or cDNA sequences encoding PRPP amidotransferases of other organisms. This DNA fragment also conferred the increased activity of PRPP amidotransferase to the host cell. Therefore the ORF was designated as purF. When this DNA fragment was expressed in the IMP-producing host cells, IPR-1, IMP production was increased due to the increased specific activity of PRPP amidotransferase. Putative start codon was decided by the analysis of E. coli promoter-like sequence. The resulting purF has a GTG start codon and codes for 53.67 kDa protein chain having 499 amino acid residues. The purF gene contains $NH_2$-terminal propeptide sequence of 24 amino acid residues and four conserved cysteine residues which participate in 4Fe-4S center causing a $O_2$-dependent inactivation in B. subtilis. The purF gene has 54.87% of G + C contents and shows a significant bias in codon utilization toward those triplet having G or C in the third position. The upstream putative promoter sequence of purF structural region matches well with the consensus promoter sequence of corynebacteria. By deletion analysis, the purF gene itself was proved to be responsible for MGS-resistance. The wild-type purF gene was cloned by the PCR method using the chromosomal DNA extracted from strain Br. ammoniagenes ATCC6872 as a template. The nucleotide sequences of wild- type purF gene was determined and compared with that of the MGS-resistant purF gene. Transitional mutations at two points, 3938 G and 4421 G of the wild type sequence to 3938 A and 4421 A, respectively, were identified. These changes resulted in amino acid replacements, Asp70Asn for nucleotide 3938 and Gly231Ser for nucleotide 4421(numbering from putative start codon). By the site-directed mutagenesis of the MGS resistant sequence to the wild type sequence, the change at position 4421 was proved to be the major cause of MGS-resistance. And when both nucleotides 3938 A and 4421 A were changed to 3938 G and 4421 G simultaneously, the MGS-resistance of the transformant was reduced to the same level of the wild type. These changes caused the release of PRPP amidotransferase from feedback inhibition by GMP.
퓨린 뉴클레오티드 생합성 경로는 5'-phosphoribosyl-1'-pyrophosphate (PRPP)로부터 10 단계의 반응을 통해 IMP가 만들어지고, 이 IMP는 각각 2 단계를 거쳐 AMP와 GMP를 합성하는 단계로 구성된다. 이는 RNA와 DNA의 전구 물질들을 생성하는 과정으로서 모든 생명체에 있어서 가장 중추적인 대사 과정이다. 박테리아의 퓨린 뉴클레오티드 생합성 경로에 관여하는 유전자들에 관해서는 대장균, 살모넬라, 및 고초균 등에서 잘 연구되었으나, 그람 양성 균이며, 비병원성이고, 간상이며, 산업적으로 정미성 퓨린 뉴클레오티드인 IMP와 GMP의 생산에 사용되는 Brevibacterium ammoniagenes 균주에서는 지금까지 이에 관한 연구 보고가 거의 없는 상태이다. 따라서 이들 퓨린 뉴클레오티드 생산 균주에 도입되는 아나로그 내성의 기작을 보다 구체적으로 밝히고 또한 퓨린 생합성 경로의 주요 조절부위 효소인 PRPP amidotransferase를 강화하여 균주의 생산성을 높이고자 하는 목적으로 그 유전자를 클로닝 하고자 하였다.
아나로그 내성을 위해서 6-mercaptoguanosine (MGS) 을 사용하였는데, 이는 유사저해제(pseudofeedback inhibitor)로서, cell 내에서 그의 뉴클레오티드의 형태로 전환되어 퓨린 생합성을 강하게 저해한다. MGS을 뉴클레오티드 형태로 전환시키지 못하는 변이에 의해서도 MGS에 대한 내성을 나타낼 수 있다. Br. ammoniagenes균주는 MGS을 뉴클레오티드 형태로 전환시킬 수 있다고 보고된 바 있다. 따라서 MGS내성 도입에 의해 IMP생산 균주의 생산성이 증가하였다면, 이 균주로부터 MGS 내성을 부여하는 DNA절편을 클로닝함으로써 PRPP amidotrans[한종권1]ferase 의 유전자를 클로닝할 수 있을 것이다. 이러한 가정하에 아래의 실험들을 진행하여 Br. ammoniagenes균주의 PRPP amidotransferase 유전자를 클로닝하였으며, MGS 내성요인을 분자level에서 규명하였다.
IMP 고생산 균주인 Br. ammoniagenes IPR-1 균주는 모균주인 IP 균주에 MGS 내성을 도입하여 제작되었으며, IMP생산성이 모균주에 비해 증가하였다. IPR-1균주의 염색체 DNA로부터 야생균주인 Br. ammoniagenes ATCC6872 균주에 MGS 내성을 부여하는 5.2kb 길이의 DNA절편을 클로닝하고 그의 염기서열을 결정하였다. 이 DNA 절편 내에서 여러 ORF들이 발견되었으며, 그 중 하나가 보고된 다른 PRPP amidotransferase 의 유전자들과 높은 유사성을 보였다. 이 DNA 절편을 도입하였을 때, 숙주세포의 PRPP amidotransferase효소 역가(specific activity)가 증가됨을 확인 할 수 있었으며, 따라서 규명된 ORF를 purF유전자라고 명명하였다. 대장균의 promoter-like sequence의 분석에 의해 promoter와 개시 코돈 부위를 결정하였다. purF 유전자는 GTG를 개시코돈으로 사용하며, 499 개의 아미노산 잔기를 갖는 53.67 kDa의 단백질을 코딩한다. Br. ammoniagenes의 PRPP amidotransferase는 고초균의 PRPP amidotransferase와 아미노산 서열상 유사한 특징을 갖는다. 즉, glutamine-dependent activity 에 관여하는 conserved active site cysteine 앞부분에 24개의 아미노산 잔기를 propeptide로 가지며, 고초균에서 효소의 산소에 의한 불활성화에 관여하는 것으로 알려진 4Fe-4S center의 형성에 관계하는 4 개의 cysteine잔기가 보존되어 있었다. 이 purF 유전자는 54.87%의 G + C content를 갖고, 코돈 triplet의 세번째 위치에서 G 혹은 C를 갖는 경향이 두드러지며 이는 다른 코리네형 세균의 유전자에서 보고된 경향과 일치한다. 또한 purF 구조 유전자 상부의 promoter서열은 코리네형 세균의 consensus promoter와 높은 유사성을 보였다. MGS 내성 기작을 구체적으로 밝히기 위해 deletion analysis를 수행한 결과, purF 유전자가 MGS 내성에 관여함을 확인할 수 있었다. PCR 방법에 의해 야생균주인 Br. ammoniagenes ATCC6872 균주의 염색체DNA로부터 야생형 purF 유전자를 클로닝하고 그의 염기서열을 결정하였다. 야생형과 MGS 내성의 purF 유전자의 염기서열을 비교한 결과, 3938과 4421의 두 위치(전체 염기서열번호)에서 야생형의 G가 A로 바뀌는 transitional mutation이 일어났으며, 결과로 3938 의 변화에 의해Asp70Asn, 4421의 변화에 의해 Gly231Ser의 아미노산 변화를 수반하였다 (putative 개시코돈으로부터의 번호). Site-directed mutagenesis에 의해 4421위치에서의 변화가 MGS내성의 주요인임을 확인할 수 있었으며, 효소역가 분석에 의해 PRPP amidotransferase가 GMP에 의한 되먹임저해(feedback inhibition) 로 부터 해제되었음을 확인하였다. 이상의 결과들로부터 IMP 고생산 균주로부터 MGS 내성의 cloning에 의해 purF 클로닝 할 수 있었으며, MGS 내성이 PRPP amidotransferase의 GMP에 의한 되먹임저해로부터 해제됨에 기인한다는 것을 보고하는 바이다.
